Semaglutide dosage and reconstitution
A weekly shot that quiets appetite; the best-proven weight peptide here. The job is keeping the muscle while the fat comes off.
Staying safe
Things people sometimes notice
- Nausea for a few days after each step up, worst on the first dose at a new level, usually gone within a month at a stable dose
- Constipation, which outlasts the nausea and needs water and fiber, or loose stools for some
- Feeling full after very little, sometimes to the point of skipping meals, which is where muscle loss starts
- Tiredness in the first weeks, often from eating too little, and sulfur burps or reflux for some
Stop and get help if (trouble breathing or swelling: call 911)
- Severe, constant stomach pain that may spread to the back, with or without vomiting, which can mean pancreatitis
- Vomiting you cannot keep ahead of, or signs of dehydration (dizziness on standing, dark urine, no tears)
- Pain in the upper right belly, especially after fatty food, which can be gallstones; rapid weight loss raises the risk
- Rash, hives, swelling of the face or throat, or trouble breathing
Talk to your doctor first, especially if
- You have had pancreatitis, or a personal or family history of medullary thyroid cancer or MEN2, because the drug is contraindicated in those cases
- You take insulin or a sulfonylurea, since your blood sugar can drop too low once semaglutide starts working and those doses usually need to come down
- You have diabetic eye disease (retinopathy), which can temporarily worsen when blood sugar improves fast
- You are pregnant, nursing or planning to be; it should be stopped about two months before trying to conceive
Mixing it
Many people get this as a prepared pen from a pharmacy. If you have a vial, use the calculator with your exact strength. Dosing is once weekly and stepped up slowly.
If prescribed, follow your pharmacy's pen instructions exactly.
If using a vial, mix per your strength and confirm the math with the calculator.
Inject once a week, same day each week.
Step the dose up slowly to limit nausea.
Store per the label, usually refrigerated.
