PT-141 + Melanotan II
Melanotan II for a tan and baseline libido over weeks, PT-141 for desire on the night. Respect the nausea, blood pressure, and your moles.
What it is
This pairing covers both ends of the melanocortin story: Melanotan II run daily for a few weeks gives you a tan and a steady lift in sex drive, and PT-141 taken an hour or two before intimacy spikes desire on demand. Both work through the brain rather than blood flow, both cause nausea and flushing until you learn your dose, and both deserve a low first shot.
The people who buy both are usually couples where desire, not plumbing, is the problem, and someone who also wants a tan without burning. Women are the group PT-141 is actually approved for (as Vyleesi, for low desire before menopause), and it works in men too. Melanotan II is the choice when you want the tan and are happy to take the libido and appetite drop that come with it. If erections are the issue, a prescription PDE5 drug is better proven and cheaper. If you want libido without the tan, PT-141 alone is the cleaner buy. If you want the tan with fewer side effects and no libido push, Melanotan I.
Timelines are different for each vial, which is the whole reason they are two vials. Melanotan II: the first few doses bring flushing and queasiness for 20 to 60 minutes, and sometimes a spontaneous erection in men; by week two the skin darkens, faster with a little sun, and libido climbs as a background hum. By week four to eight you hold the tan at two or three shots a week. PT-141: nothing for 45 minutes, then a flush, often nausea, and over the next two to six hours a noticeable rise in desire and arousal that is not tied to a partner doing anything. It lasts into the next day for some people. Neither does anything for someone whose problem is pain, a relationship, or a medication side effect.
Run them on their own schedules and do not combine them until you know each. Melanotan II: 250 to 500 mcg subQ (just under the skin with an insulin needle) once a day in the evening so you sleep through the nausea, for four to eight weeks, then two or three times a week to hold. Our advice is a 100 mcg test shot the first night before stepping up. PT-141: 200 mcg the first time, 500 mcg as the usual dose, and up to 1 mg only if 500 was clearly not enough, 45 minutes to two hours before, never more than one shot in 24 hours and no more than eight a month. Nausea is dose-driven, which is why the first shot is small; the approved Vyleesi pen is 1.75 mg, and we stop at 1 mg. Mix each 10 mg vial with about 3 ml of water. A 10 mg PT-141 vial is twenty doses at 500 mcg; a 10 mg Melanotan II vial is 20 daily loading shots at 500 mcg or 40 at 250 mcg, so a longer load needs a second vial. The mistakes: dosing both the same evening the first week, taking PT-141 on a full stomach (it works, you just feel worse), and skipping the photo of your moles.
The evidence is split. PT-141 is a real approved drug: two large trials in women with low desire showed a modest but real improvement in desire scores against placebo, with nausea in about four in ten and most people adapting after a few doses. Melanotan II never finished development; it has small human studies from the 1990s on tanning and on erections in men, then went straight to the gray market. What is well documented for both is the blood pressure bump after each dose (the reason for the eight-a-month limit) and, for Melanotan II, darkening of moles and freckles and reports of new ones, which is why anyone with a lot of moles or a melanoma history should skip it.
Not worth it if you only need one of the two: PT-141 alone for desire, Melanotan I alone for a gentler tan. Oxytocin is the alternative when the gap is connection and warmth rather than drive. Skip the pair entirely with uncontrolled blood pressure, heart disease, a melanoma history, or a lot of moles; and if you are a man prone to prolonged erections, Melanotan II is the one to leave out.
What people use it for
Desire and arousal
Some evidencePT-141 (bremelanotide) was approved as Vyleesi in 2019 after two large trials in premenopausal women with low desire showed a modest but statistically real gain in desire scores over placebo. Smaller earlier studies showed effects in men. It works through brain melanocortin receptors, not blood flow, so it helps when wanting is the problem and does nothing for mechanics.
Tanning and sun tolerance
Some evidenceSmall 1990s human studies showed Melanotan II darkens skin with little or no sun and that both melanotans reduce burning. It never finished development, so there is no large trial and no long-term safety data. The tan is real and predictable; the pigment also lands on moles and freckles, which is the trade.
Erections
Early daysMelanotan II produced erections in small studies of men with erectile dysfunction in the 1990s, which is where the PT-141 program came from. The effect is real but unreliable and comes with the full nausea and flushing package. For erections specifically, PDE5 drugs are better proven and far more predictable.
How people use it
Safety, in brief
Things people sometimes notice
- Nausea and a hot flush for 20 to 60 minutes after a dose, worse at higher doses and on a full stomach; most people adapt after a few shots
- A general darkening of skin from Melanotan II, including freckles, moles, and sometimes gums or lips
- Less appetite for a day after a Melanotan II shot
- Spontaneous erections in men in the hours after either peptide, especially early on
