MitoStack (SS-31 + MOTS-c)

SS-31 and MOTS-c in one vial: the mitochondrial oil change plus the gas pedal. Subtle energy and endurance over weeks, not a stimulant.

What it is

MitoStack puts SS-31 and MOTS-c together for steadier energy, better exercise tolerance, and a mitochondrial tune-up in one daily shot. Think of SS-31 as the oil change that makes the engine run clean and MOTS-c as the gas pedal that tells it to burn fuel; what you feel is subtle and builds over four to eight weeks.

The person reaching for this is usually in their forties or fifties and has noticed that stamina is the thing slipping: workouts that used to feel fine now cost two days, afternoon energy craters, recovery from a hard week takes a week. A second group is endurance athletes and lifters who want more capacity in a training block. Many have already run NAD+ and want the next layer. It is not for anyone expecting a caffeine-like lift, and it is the wrong tool for fat loss on its own; MOTS-c nudges metabolism, it does not suppress appetite.

What you notice, if you notice anything, is steadier: fewer afternoon dips in the first one to two weeks, then over weeks three to six workouts feel more sustainable and you recover faster between them. Some people feel nothing at all, and that is a common outcome with mitochondrial peptides. Good looks like an extra interval, a lower resting heart rate on a tracker, or simply a week that did not flatten you. It will not build muscle, it will not burn fat on its own, and it will not fix sleep.

The standard rung is 4 mg total, which is 2 mg of each, subQ (just under the skin with an insulin needle) once a day for four to eight weeks, then a few weeks off. Neither peptide is known to stop working with use; the break is caution, since there is little human data on running them together for long. The gentle rung is 2 mg total daily; the higher rung is 6 mg three times a week, which is closer to how MOTS-c is usually pulsed. Morning dosing suits most people since MOTS-c can feel slightly activating. The ratio is the catch: the vial is a fixed 1:1, so at 4 mg you get a normal SS-31 dose and about double the usual daily MOTS-c dose (1 mg is the solo standard). If that bothers you, run the gentle rung or buy the two peptides separately. Mixed at 2.5 ml, a 50 mg vial is 12 doses at 4 mg, so an eight week daily run is four or five vials; keep it out of light, as with most peptides. The mistakes: starting NAD+ the same week and never learning which one helped, and judging it at day ten.

The evidence splits down the middle. SS-31, as elamipretide, has real human trials in mitochondrial myopathy, Barth syndrome, and heart failure, with some functional improvements and some trials that missed their main endpoint; it is the best-studied half and the only mitochondrial peptide here with human data. MOTS-c improved insulin sensitivity and running capacity in rodents and tracks with fitness in human blood samples, with no human dosing trials. Nobody has tested the two together, and the 1:1 ratio is a packaging decision, not a finding. Promising but not proven, with the SS-31 half ahead of the MOTS-c half.

If you only want one, SS-31 alone is the one with human data, and MOTS-c alone is cheaper and lets you dose it 1 mg daily or 3 mg a few times a week the normal way. NAD+ is the simpler first step for general energy, with a lot more user experience behind it. HNG (humanin) is the other mitochondrial-derived peptide for people who want to go further down this road. Skip MitoStack if you take insulin or a sulfonylurea without a plan for the glucose drop MOTS-c can cause.

What people use it for

Cell energy and muscle function

Some evidence

SS-31 (elamipretide) has been through human trials in primary mitochondrial myopathy, Barth syndrome, and heart failure: some showed better walking distance or heart measures, others missed their main goal. Animal data on protecting the inner mitochondrial membrane is consistent. It is the grounded half of the blend, studied in sick patients, not healthy adults.

Metabolism and endurance

Early days

MOTS-c improved insulin sensitivity, fat handling, and treadmill running capacity in mice, including old mice, and its blood levels rise with exercise in people. There are no human dosing trials. For you that means a plausible endurance and metabolic nudge with the dose borrowed from animal work.

The two together

Early days

No study has combined SS-31 and MOTS-c. The case is mechanistic: one stabilizes the membrane where energy is made, the other signals the cell to burn fuel and build more mitochondria. The 1:1 vial ratio is a convenience, and it pushes MOTS-c above its usual daily dose at the standard rung.

Safety, in brief

Things people sometimes notice

  • Redness or itching where you inject, gone in a day
  • A headache or flushing in the first few days
  • Feeling slightly wired, or slightly flat, in week one while your body adjusts
  • Lower blood sugar readings if you check; MOTS-c improves insulin sensitivity
